This particular area isn’t necessarily locked, as I recall; you don’t strictly have to complete the related quest to gain access. However, to proceed deeper into the Underground Lava River, it’s essential that you initiate the quest [Tale of Dreams Plucked From Fire Quest Chain], which is the same one from the previous pathway.
But to get to the Sprayfeather Gill, it’s hidden behind a simple puzzle (unrelated to the quest). Approach this seelie-looking creature (called Monetoo in natlan) and it will go back inside its mural. Infuse the mechanism with Phlogiston and the hidden door will open.
