It’s common for superheroes to have weak or unimportant villains when they first appear in comics, with only one or two eventually becoming significant. Usually, it takes years for a hero to develop a true arch-enemy. But Spider-Man was different. In his first year of comics, an incredible six of his most famous villains were introduced – names still associated with him today. Plus, three more major foes – The Green Goblin, Mysterio, and Kraven the Hunter – debuted near the end of that first year, quickly establishing one of the most impressive groups of villains in all of comics.
Here’s a ranking of the first ten supervillains Spider-Man ever battled in comic books, from iconic foes like Doctor Octopus and Sandman to some lesser-known ones, judged by how memorable they’ve become over time.
10) The Enforcers

It’s amusing to read the old tagline for The Amazing Spider-Man #10, which boasted that no villains were as ruthless as the Enforcers. However, they quickly faded into the background after Spider-Man faced the much more formidable Sinister Six. The Enforcers were a trio – Montana, Fancy Dan, and the Ox – who worked for the crime boss known as Big Man. While they appeared in 48 comics fighting Spider-Man or Daredevil, they were always just minor, run-of-the-mill bad guys.
9) Living Brain

The Living Brain first appeared in The Amazing Spider-Man #8 as a robot created by Dr. Petty. It was brought to Peter Parker’s school as a demonstration of advanced problem-solving abilities. However, a malfunction caused it to go out of control, and Spider-Man had to stop it. This story was notable for introducing Peter’s teacher, Raymond Warren, who was the brother of the villain Miles Warren, also known as the Jackal. The Living Brain reappeared on occasion over the years, even joining the Sinister Six, and has a total of 41 appearances in Marvel comics.
8) Tinkerer

Phineas Mason, known as The Tinkerer, first appeared in Amazing Spider-Man #2, making him one of Spider-Man’s earliest foes. His initial plan involved hiring actors to stage a fake alien invasion as a distraction, allowing him to steal classified government information. However, his scheme was quickly exposed, and Spider-Man helped capture him. Notably, he was initially employed by Quentin Beck, who would later become the supervillain Mysterio, though Mysterio didn’t appear in the comics until a later date. The Tinkerer proved to be a recurring character, appearing in a total of 68 issues.
7) Chameleon

Interestingly, the very first villain Spider-Man faced was the Chameleon. His debut in The Amazing Spider-Man #1 featured two stories: one where he attempted to join the Fantastic Four, and another where he battled the Chameleon, a skilled master of disguise. The Chameleon took advantage of Spider-Man being a wanted man, disguising himself as the hero to commit a robbery. Spider-Man stopped him and proved his own innocence. The Chameleon later became a key figure by introducing his brother, Kraven the Hunter. Over the years, he’s appeared in an impressive 175 issues of Marvel Comics.
6) Electro

Electro first appeared in Amazing Spider-Man #9. Originally an electrical technician, he developed superpowers after an accident and quickly chose a life of crime. He initially fought Daredevil before refocusing his efforts on Spider-Man. Realizing he needed help to defeat Spider-Man, Electro frequently joined forces with the Sinister Six. Over time, his powers grew, eventually transforming him into almost pure energy, but at the cost of his humanity. Throughout his comic book history, Electro has appeared in a total of 221 issues.
5) Vulture

The Vulture first appeared in The Amazing Spider-Man #2 as Spider-Man’s first super-powered foe – though his abilities came from technology, specifically a flight suit. Initially, the Vulture was just a common thief, and Spider-Man had to rely on his intelligence to defeat him by disabling the villain’s equipment. He also became the first recurring villain for Spider-Man, reappearing in Amazing Spider-Man #7. Over the years, the Vulture has become one of Spider-Man’s most recognizable enemies, appearing in a total of 233 issues.
4) Lizard

Early in his career, Spider-Man faced many villains, but Curt Connors was perhaps the most heartbreaking. A well-meaning scientist and war veteran who lost a hand, Connors tried to regrow it using reptile DNA. Unfortunately, the experiment transformed him into a massive, uncontrollable lizard – a tragic twist on the Jekyll and Hyde story. This transformation made Spider-Man determined to cure Connors instead of simply defeating him. Over the years, The Lizard’s story has become quite complex, and he’s appeared in a total of 215 comic books.
3) Sandman

Flint Marko, known as Sandman, first appeared in Amazing Spider-Man #4 and is a classic Spider-Man villain. He was unique as the first of Spider-Man’s foes to have powers instead of relying on gadgets. After an accident involving radiation, Sandman gained the ability to transform into sand, making him a difficult opponent. Over the years, he’s consistently challenged Spider-Man, both on his own and as a member of the Sinister Six. Interestingly, Sandman has also tried to reform, becoming friends with The Thing and even briefly joining the Avengers. He’s been featured in a total of 312 comic book issues since his debut.
2) Doctor Doom

Surprisingly, Doctor Doom was only the fifth villain Spider-Man ever faced! This happened in The Amazing Spider-Man #5, and it was definitely the real Doctor Doom – not one of his robotic duplicates. Though Doom first appeared the year before in Fantastic Four #5 as a foe of that team, here he tried to recruit Spider-Man to help him fight the Fantastic Four. Unsurprisingly, Spider-Man said no, leading to a battle where the Fantastic Four had to rescue him. While Doctor Doom has appeared in 979 comics overall, he’s not primarily a Spider-Man villain, which puts him at number two on the list.
1) Doctor Octopus

Doctor Octopus stands out as the most memorable villain Spider-Man has ever faced. First appearing in Amazing Spider-Man #3, he quickly became Spidey’s greatest foe, even surpassing the Green Goblin in notoriety. He’s one of the few villains to actually win against Spider-Man, famously taking his place for a significant period in the Superior Spider-Man storyline. With over 555 comic book appearances since his debut, Doctor Octopus continues to be Spider-Man’s most iconic and enduring enemy.
